제목 Hierarchical topography enhancing cardiomyocytes maturation through synergistic stimulation of nano-and micro-structure
초록 Recently, the topography of biomaterials has provided an essential scaffold mimicking the niche environment. Remarkable improvements have been achieved to provide biophysical cues in micro and nano-scale, but there still remain challenges for mimicking multiscale hierarchical features of native tissues. Here, we developed the hierarchical topography combining micro-wrinkle (μW) and nano-pillar (nP). First, we observed the response of cardiomyocyte (CM) depending on the wavelength of uW, which showed two types of cellular alignment. Also, CMs were tightly attached on the low aspect ratio nPs (A.R < 0.8) and formed focal adhesion densely. Through systematical investigation, we fabricated the hierarchical structure incorporating μW and nP. On the hierarchical structures with large groove (cell width < wavelength), we found the synergistic stimulation for the cardiac maturation of H9C2 and iPSC-derived cardiomyocytes. Confined within groove, CMs could form focal adhesion intensely and advance the maturation (~ 3 times higher). This topographic effect was mediated by lamin expression. We believe this systematically designed hierarchical topography can provide new insights for multi-scale biophysical stimulation platforms.
