제목 | Engineering of the substrate binding pockets of α-ketoglutaric semialdehyde dehydrogenase for enhancing the activity with regard to 3-hydroxypropionaldeyhde |
초록 | 3-Hydroxypropionic acid (3-HP) is a key building block for valuablechemicals. A biological route for synthesizing this molecule is two-step enzymatic reactions; dehydration of glycerol to 3-hydroxypropionaldehyde (3-HPA)by glycerol dehydratase and then oxidation of 3-HPA to 3-HP by aldehydedehydrogenase. Here, we report an aldehyde dehydrogenase, anengineered a-ketoglutaric semialdehyde dehydrogenase from Azospirillumbrasilense. The variant, named 2C10, was obtained by applying alarge ALDH library to a transcription factor based selectionmethod and then a screening method for observing the activities ofindividual clones. 2C10 exhibited a 4.65-fold higher catalyticactivity toward 3-HPA than the WT. The flask culture of Pseudomonasdenitrificans with 2C10 resulted in an approximately 30% increasein 3-HP titer compared with that obtained using WT. MD simulationssuggested that compared to the WT, 2C10 has a less flexible and smaller bindingpocket for aldehyde substrates. |
