제목 An Injectable Anti-inflammatory Hyaluronic Acid Hydrogel with Decellularized Brain ECM and TI-EVs using Neural progenitor cells for Spinal cord Regeneration
초록 Traumatic spinal cord injury (SCI) is a serious condition that can cause permanent motor dysfunction including neurological dysfunctions and pain. In this study, we fabricated a hyaluronic acid (HA)-based hydrogel combined with decellularized brain matrix (DBM), polydeoxyribonucleotide (PDRN), TNF-α/IFN-γ primed mesenchymal stem cell-derived extracellular vesicles (TI-EV), and human neural progenitor cells (NPCs). This hydrogel (DBM/PDRN/TI-EV/NPC in HA gel) could supply structural support to nerves, and we predicted that nerve regeneration would be possible due to the synergistic effects of the included substances. The results showed that the hydrogel enhanced the proliferation and differentiation of NPCs in vitro. Functional motor recovery was also confirmed to be possible by reducing the inflammatory response and forming neurons and oligodendrocytes in vivo animal tests. This new bioactive injectable HA-based hydrogel is an effective candidate for SCI regeneration.
