제목 DNA cancer vaccine empowered by an injectable mesoporous silica microrods scaffold for cancer immunotherapy
초록 DNA vaccine is one of the promising approaches in immunotherapy for the treatment of cancers. To induce an effective antigen-specific adaptive immune response, the DNAs coding the antigen are desired to be delivered into antigen-presenting cells (e.g., dendritic cells (DCs)); however, the effective delivery of DNAs to those cells is still limited. Here we propose an injectable porous scaffold to empower the efficacy of DNA vaccine by augmenting the DNA delivery to host DCs. Mesoporous silica rods (MSRs) were loaded with chemoattractant for the DCs and antigen-coded DNA-polyplex to prepare MSR-DNA vaccine. Subcutaneous injection of MSR-DNA vaccine resulted in the recruitment of a significant number of DCs, a high cellular uptake of antigen-coded DNA by the recruited DCs, and more robust host DCs activation and antigen-specific CD8+ T cells compared to the bolus DNA vaccine counterpart. Impressively, the vaccine significantly inhibited established lung metastasis with a single injection. Our results indicate that MSRs could serve as a powerful delivery platform of DNA vaccine to DCs for an effective cancer immunotherapy.
