一方で、天皇を中心とした新しい政権を打ち立てるべきであるとする尊王攘夷派の公家等と公武合体を支持する公家が抗争を始めた。佐幕派の公家と三条実美と岩倉具視を中心とした尊王攘夷側の公家の間で抗争が生まれ幕府大老井伊直弼による安政の大獄はそのような状況の中で行われた。
幕末時において、徳川幕府による政権体制を認めていた孝明天皇(攘夷の思想を支持)が若く亡くなり。天皇が将軍家として大政を委ねられていた徳川幕府は、一時は公武合体を目論んで、天皇に対して大政奉還をすることを決断し、実行したが、天皇を御旗とした新しい政権を打ち立てるべきであると主張する尊王攘夷の公家等は公武合体の体制を認めず徳川幕府を討伐・追放する流れになった。
その戦いの節目の戦いが、京都の伏見の戦いであった。長崎の出島に駐留していた英国商人グラバーによって手配された西洋の近代的な大砲、鉄砲等の兵器を収受した後に、天皇の錦の御旗を掲げた尊王攘夷の倒幕の軍隊は徳川幕府軍を倒して、明治維新を迎えることになった。
そして、尊王攘夷派の公家三条実美、岩倉具視の中心とした主導で、孝明天皇の後継であった幼少の明治天皇を錦の御旗に祀り上げ、明治天皇の基で、尊王攘夷の思想概念に基づいた明治維新政府が開始された。

嘉永元年(1848年)に武家伝奏となり、たびたび江戸に下って対米政策について江戸幕府と交渉した。安政5年(1858年)には日米修好通商条約への勅許を巡り関白・九条尚忠と対立して、3月7日には左大臣・近衛忠煕と共に参内停止を命じられる。これに激怒した孝明天皇によって2日後に右大臣・鷹司輔煕と権大納言・二条斉敬を勅使として近衛・三条両邸に派遣して両名に参内の勅命を下した。これは長年朝廷の全権を握っていた摂関家が勅使となり、政治的に非力であった清華家出身の三条を出仕させるという公家社会始まって以来の出来事として衝撃を与えた。
安政6年(1859年)4月に安政の大獄で謹慎の処分を受け、出家して澹空と号した。同年、病気危篤となり、従一位に叙され、10月に謹慎していた一条寺村で薨去。薨後の文久2年(1862年)に右大臣が追贈され、明治2年(1869年)には明治天皇より「忠成公」の諡を与えた。さらに、1899年(明治32年)には贈正一位に追陞された。

徳川幕府の将軍徳川家斉(1773年11月18日〜1841年2月27日)の時代に大奥で風紀の紊乱が行われていた。徳川家斉も50数人の子供をつくり、諸藩に与え、諸藩の憤りを被っていた。徳川家斉は、“江戸の300年にこれほどめでたい人はいない”と後世にいわれている。水野忠邦は日蓮宗感応寺の僧と大奥女官との怪しげな関係の調査を寺社奉行阿部正弘に指示し、阿部が高級女官30余人を吟味した大奥の調査で、彼女たちと僧日啓、日量(お美代方実兄)。日尚(同甥)らとの淫らな関係がわかった。感応寺はとり潰し。幕末において幕府本体の機能が腐敗し、崩壊に向かっていた*。

徳川 家斉(とくがわ いえなり)は、江戸幕府の第11代征夷大将軍(在任:1787年 - 1837年)。
御三卿一橋家の第2代当主徳川治済の長男。母は側室のお富の方。
寛政元年(1789年)、島津重豪の娘・近衛寔子と結婚している。

幕末のオランダ人のシーボルトは伊能忠敬が調査して作り上げた機密の日本地図を手に入れて、国外追放処分になった。
間宮林蔵がシーボルトが怪しいと代官に訴えていた。そのために、捜索をされたのでしょう。
シーボルトは国内で取り調べを受けて、国外追放の処分となった。日本地図は、没収されたはずであるが、シーボルトに対する日本人の影の協力または、別の場所に隠していた日本地図を、捜査を逃れて、国外に持ち出したのでしょう。
没収された日本地図を再度手に入れたのかあるいはどこかに隠していたのかは不明であるが、海外に持ち帰っていて、日本に開国の交渉をする前に米国のペルー等に伝わっていた。
シーボルトは日本の鎖国を開国するために使用するとの米国の意向の説明を受けて、その依頼に対して、軍事的な利用をしないという条件を米国側につけて、シーボルトは日本の地図を米国へ引き渡した。
このシーボルトが本国オランダに持ち帰った日本地図は、すぐに米国に伝わった理由を究察することも興味あることです。たぶん、オランダのキリスト教の宣教者が米国に伝えたのでしょう。?
その後、シーボルトの子孫も来日して、日本で暮らすことになる。
言い換えれば、鎖国状で機密戦略上重要な日本の地図を、すでに米国のペルー側が利用していた。用意周到にして、来日したことが理解される。
米国のペルーは、その約束を守り、徳川幕府に日米の交渉に当たったことが、日米和親条約の成文の中で、謳われている。
補追:
シーボルトは博学者で、研究のため日本の植物や動物を収集していた。医者としての知識も持ち合わせしていた。
日本のオオサンショウウオもオランダに持ち帰っている。当地で死んだオオサンショウウオの標本が博物としてオランダのライデン博物館に収納されているとのことです。
