The San Francisco 49ers will enter Week 1 without an important piece of their defensive line. Defensive lineman Sam Okuayinonu has been ruled out against the Los Angeles Rams .
Okuayinonu, who registered 39 tackles, 23 solo tackles, six tackles for loss, three sacks, and one forced fumble in 15 games, was initially listed as questionable with a foot injury. However, the Niners recently downgraded him to out before the regular-season opener.

NFL analyst Matt Maiocco reported the latest development as the 49ers prepared for their trip to Australia via his X handle. Okuayinonu did not travel with the 49ers to Melbourne for the Week 1 matchup. San Francisco departed for Australia on Wednesday night ahead of the international contest.
The 49ers will face the Rams at Melbourne Cricket Ground on Thursday, September 10. The game represents one of the NFL's unusual opening-week divisional matchups. Okuayinonu's absence creates another concern for a defensive line already dealing with limited depth. Nick Bosa traveled with the team but is returning from his third ACL injury.
San Francisco also placed 2025 first-round pick Mykel Williams beginning the season on reserve/PUP. The applicable rule means he cannot return until after the first four games. That leaves Keion White and 2026 third-round pick Romello Height as the other primary defensive ends potentially available. Although Height has also dealt with an injury, White is listed as questionable for Week 1, they have a higher chance of featuring on the roster than Okuayinonu.
White could receive the starting opportunity opposite Bosa. Height could then provide additional pressure in passing situations. The 49ers could also elevate a practice-squad player before kickoff.
The interior defensive line offers San Francisco more depth. Osa Odighizuwa , who joined San Francisco from the Dallas Cowboys for a 2026 third-round pick, should handle one starting tackle position after joining the team through an offseason trade. Alfred Collins and C.J. West also remain options inside.
Yet, there's no denying that Okuayinonu's absence still represents a notable loss because of his experience. The 28-year-old is entering his fourth NFL season and third campaign with San Francisco.
He became especially important last season after injuries hit the 49ers' defensive end group. Bosa and Williams both suffered season-ending injuries during the 2025 campaign. The 28-year-old appeared in 15 regular-season games and made 12 starts. He finished the season with three sacks and six tackles for loss. Okuayinonu also played all 16 games in 2024. His experience gives the Niners a reliable rotational presence that the team will now miss against the Rams
Elsewhere, Bosa's workload will require careful management following his ACL recovery. So, chances are high the 49ers cannot assume their star pass rusher will immediately handle his previous workload. It's also why we could see White face increased responsibility in the opener. Similarly, Height may receive an expanded role depending on how San Francisco manages its pass-rush packages.
