The Danish biotech company reported positive topline results from its Phase 3 LiGeR-HN1 trial, which evaluated petosemtamab in combination with pembrolizumab versus pembrolizumab alone in patients with first-line PD-L1 positive recurrent/metastatic head and neck squamous cell carcinoma.

The trial met both primary endpoints at its first interim analysis, showing a statistically significant improvement in overall survival and objective response rate. The combination treatment reduced the risk of death by 50% compared to pembrolizumab alone, with an overall survival hazard ratio of 0.50.

The safety profile of petosemtamab combined with pembrolizumab was generally well tolerated and consistent with previously reported safety profiles of the individual agents. No new safety signals were identified.

Petosemtamab is an investigational IgG1-bispecific antibody targeting the epidermal growth factor receptor and the leucine-rich repeat containing G-protein-coupled receptor 5.

Genmab plans to engage global regulatory authorities and will submit full results for presentation at a future medical meeting.

Beyond LiGeR-HN1, petosemtamab is being evaluated in multiple Phase 3 trials, including LiGeR-HN2 in patients with second- and third-line recurrent/metastatic head and neck squamous cell carcinoma, and two Phase 3 studies in patients with previously treated colorectal cancer.
